Does anyone know if there is any HDB rule for second-timer BTO, stating that they must upgrade/downgrade from the first BTO size?

Heard rumours saying that for second timer BTO, there's a rule saying must upgrade from first BTO size and cannot downgrade.

There is no such rule, but there is a re-sale levy that needs to be paid if you wish to buy a 2nd BTO.
