I have a younger colleague, who is a local uni grad, whom I came to know that her salary is almost about the same as me. I have about 6 years more work-related experience than her and is holding a professional qualification. Am I in the position to request for higher salary, assuming I find a new job?

Both qualifications and experience can be important factors in determining salary, but the main factor is managing individual expectations and market rates. You will get the minimum salary unless you ask for it and is market range. I will ask depending on profitability and revenue size of the company. If the company is doing well... yes, that's best time to ask.

Agreed with what Billy said, "Networks & Connections" are more important than a paper qualification.
A paper qualification (local or overseas) is just like an entry ticket to your first job and the rest is up to your performance.
There's a saying, "can study doesn't mean can work".
Do not feel insecure and infurious about this. You can continue to send out more resumes while you think of a game plan to negotiate with your boss.
If your boss hesitated or failed to give you a proper answer, you could always go find another job. There is no harm in trying.
Never try, never know.

It's the system unfortunately, the paper will be used as a baseline and prioritised, if not schools would be out of business. People look at qualifications the same way they look at Rolexs'. Lol.
I for one value skill sets and experience, but again who am I? Ask for something better if they value you, they'll probably give you, else move forward.
