Which would be better on SQ? buying an economy ticket and then upgrading it to Business class using miles; or straight up redeeming a business ticket using miles? Which is more mile-efficient?

The more mile-efficient option is to straight up redeem a business ticket using miles.
I'm not sure how true this could be across all airlines but I've a friend who often dresses up really nicely at airports. Despite being on economy class, the airline periodically upgrades her to business class without additional cost.

Not worth in 99% of scenarios. Sometimes they do a last min upgrade promos on miles and might send an email - based on how much you have paid for economy tickets.
In that case, it might make sense
