I quit my job in 2015 and took a Masters course. Then, I started working again in 2018. But I did not claim for Course Fees Relief on my IRAS Tax Filing in 2019. Can I claim the Tax Relief now (2020)?

Yes, it is possible but you have to meet certain conditions set out by IRAS. From the IRAS website:
Deferring Course Fee Relief Claims
"Your assessable income is $22,000 or below throughout the duration of the course, seminar or conference that leads to an approved academic, professional or vocational qualification and claim the relief:
In the first Year of Assessment when your assessable income exceeds $22,000; or
Within 2-YA period from the YA relating to the year you have completed the course or attended the seminar or conference, whichever is earlier."
